چکیده
Supplemental Experimental Procedures OAZ-lacZ Gene-Trap Clone XB409 The ES cell clone was obtained from Baygenomics (www. baygenomics. ucsf.edu), the site of genomic insertion and fusion of OAZ gene with lacZ was confirmed by RT-PCR from ES RNA, and injected into C57BL/6 blastocysts. The genomic insertion site was mapped by genomic southern blot and confirmed by sequencing. After germ line transmission, the heterozygous OAZ mice were backcrossed with C57BL/6, and the line was maintained either by heterozygous intercrosses or by heterozygous backcrossing to C57BL/6 mice. Homozygous OAZ pups were born, but died within one day of birth, indicating the insertion also disrupts OAZ function.

Zfp423/OAZ Participates in a Developmental Switch during Olfactory Neurogenesis
The coordination of gene expression is critical for cell differentiation and the subsequent establishment of tissue function. We show here that a multiple zinc finger transcription factor, Zfp423/OAZ, is transiently expressed in newly differentiating olfactory-receptor neurons (ORNs) and has a key role in coordinating the expression of immature and mature stage-specific genes. OAZ deletion in m...
متن کاملZfp423/OAZ mutation reveals the importance of Olf/EBF transcription activity in olfactory neuronal maturation.
Zfp423/OAZ, a multi-zinc finger protein, is proposed to participate in neuronal differentiation through interactions with the Olf/EBF (O/E) family of transcription factors and mediate extrinsic BMP signaling pathways. These activities are associated with distinct domains of the Olf/EBF-associated zinc finger (OAZ) protein. Sustained OAZ expression arrests olfactory sensory neurons (OSNs) at an ...
متن کاملOAZ Uses Distinct DNA- and Protein-Binding Zinc Fingers in Separate BMP-Smad and Olf Signaling Pathways
We have identified the 30-zinc finger protein OAZ as a DNA-binding factor that associates with Smads in response to BMP2, forming a complex that transcriptionally activates the homeobox regulator of Xenopus mesoderm and neural development, Xvent-2. OAZ contains a BMP signaling module formed by two clusters of fingers that bind Smads and the Xvent-2 BMP response element, respectively. Previously...
متن کاملFetal development of subcutaneous white adipose tissue is dependent on Zfp423
OBJECTIVE Zfp423 is a multi zinc-finger transcription factor expressed in preadipocytes and mature adipocytes in vivo. Our recent work has revealed a critical role for Zfp423 in maintaining the fate of white adipocytes in adult mice through suppression of the beige cell thermogenic gene program; loss of Zfp423 in mature adipocytes of adult mice results in a white-to-beige phenotypic switch. How...
متن کاملA developmental switch of axon targeting in the continuously regenerating mouse olfactory system.
The mammalian olfactory system has the natural capacity to regenerate throughout the animal's life span. Despite constant neurogenesis, olfactory sensory neurons project to precise, stereotypical positions in the brain. Here, we identify a critical period of olfactory sensory axon targeting during postnatal development in mouse. Perturbing axon projection beyond postnatal day 7 permanently disr...
